摘要
目的制备分泌血吸虫抗原的杂交瘤。方法采用杂交瘤技术,将血吸虫成虫细胞与Sp2/0骨髓瘤细胞融合,HAT选择性培养。结果获得了1株培养上清能与血吸虫病人血清起反应的杂交瘤,其染色体数为72~84,该杂交瘤细胞可持续培养11周以上,但其生长缓慢,在培养8周后,出现细胞逐渐死亡。结论杂交瘤技术不适于远缘杂交。
Objective To preduce the hybridoma expressing Schistosomiae antigen. Method Cells from adult Schistosoma japonicum were fused with Sp 2/0 mouse myeloma cells, and cultured with HAT media. Results A positive clone was obtained, its chromosome number was 72 ̄84. The culture supernatant gave a positive reaction with serum from patients infected with S. japonicum, as demonstrated by ELISA. The hybridoma lived in HAT media over 11 weeks. After they were cultured for 8 weeks, the cells died slowly. Conclusion Cell death may be caused by segregation of DNA and loss of chromosome material. Hybridma technique is not suitable for distant hybridization.
