反相HPLC监控G-CSF工程菌发酵过程中的乙酸含量

摘要 采用HPLC和GDAS分析测定G-CSF基因工程菌在发酵罐发酵培养过程中产生的乙酸含量。分析确定了乙酸含量与产物表达的量化关系,验证了乙酸抑制产物表达的作用。建立了HPLC测定培养液中乙酸含量的方法,该方法准确快速,检测灵敏度高,仪器分析重现性好,样品回收率高。 The content of acetic acid accumulated in the process of fermenting G-CSF genic engineering bacteria in the fermentaion tank was determined by antiphas HPLC. A relation between acetic acid content and expression of G-CSF genetically engineered bacteria was analysed and established. The G-CSF expression was inhibited by accumulation of acetic caid The method of detecting the content of caeticacid was established and it is accurate,rapid,and highly sensetive. Using this method,the recovery rate of acetic acid was high.
