细胞融合肝癌疫苗体内抗肿瘤特异性及T细胞作用的初探 被引量:2

摘要 目的:观察大鼠BERH-2肝癌细胞与自体激活B细胞的融合细胞作为疫苗抗肿瘤作用的特异性,并观察该肝癌疫苗体内抗肿瘤作用与T细胞亚群的关系。方法:(1)应用聚乙二醇(PEG)融合大鼠BERH-2肝癌细胞与激活B细胞制备细胞融合大鼠肝癌疫苗;(2)事先经该肝癌疫苗免疫的大鼠分别接种肝癌BERH-2细胞和大鼠膀胱癌NBT-Ⅱ细胞,观察该肝癌疫苗抗肿瘤作用的特异性;(3)该肝癌疫苗免疫大鼠之前或之后,以小鼠抗大鼠CD4单抗或抗CD8单抗多次注射,以清除大鼠CD4+或CD8+T细胞,随后观察大鼠的成瘤性。结果:BERH-2细胞不能在经细胞融合大鼠肝癌疫苗免疫的大鼠体内致瘤,而NBT-Ⅱ细胞可致瘤;事先清除CD4+或CD8+T细胞的大鼠经细胞融合大鼠肝癌疫苗免疫后仍能被肝癌BERH-2细胞致瘤;而经该肝癌疫苗免疫的大鼠,清除了CD4+细胞后不能被BERH-2细胞致瘤,清除了CD8+细胞后则可被BERH-2细胞致瘤。结论:细胞融合大鼠肝癌疫苗有特异性抗肿瘤作用。 Objective: To observe the special antitumor effect of the cell fusion rat hepatic tumor vaccine made by cell fusion between rat BERH 2 hepatic tumor cell line and autologous B lymphocyte and to observe the relationship between the special antitumor effect of this tumor vaccine and the subtypes of T cells. Methods: The fusion cell rat hepatic tumor vaccine was made by cell fusion between rat BERH 2 hepatic tumor cell line and activated B lymphocyte and PEG was used as the fusion agent. Two groups of rats ,immunized by the vaccine first, were then inoculated by BERH 2 cells and rat NBT Ⅱ bladder tumor cell line respectively, and then the special antitumor effect was evaluated. In order to know the relationship between the special antitumor effect of the tumor vaccine and the subtypes of T cells, the rats, either before or after its immunization with the vaccine, were injected by mouce anti rat CD4 monoclonal antibody or by mouse anti rat CD8 monoclonal antibody several times to eliminate CD4 + or CD8 + T lymphocyte correspondingly, and then, tumorigenicity was observed. Results: Rats, first immunized with the vaccine, developed no tumor after BERH 2 cells inoculation, while developed tumor after inoculated with NBT Ⅱ cells. Both groups of the rats, whatever eliminated CD4 + or CD8 + T lymphocyte, developed tumor after immunization by the tumor vaccine. Rats, first immunized by the tumor vaccine, were divided into two groups, one group, which was eliminated CD4 + T lymphocyte and then were injected with BERH 2 cells, didnt develop tumor; the other group, which was eliminated CD8 + T lymphocyte and then also injected with BERH 2 cells, did develop tumor. Conclusion: The fusion cell rat hepatic tumor vaccine has special antitumor effect, which is closely related to CD4 + and CD8 + T lymphocyte.
