健骨二仙丸提取物对大鼠体外培养成骨细胞PKC和Bag-1的影响 被引量:1

Effect of Jiangu Erxian Pill Extract on PKC and Bag-1 in Rat Osteoblast in Vitro

摘要 目的:探讨健骨二仙丸提取物对大鼠体外培养成骨细胞的作用机理。方法:采用新生大鼠颅骨进行成骨细胞分离,并在含10%的胎牛血清DMEM中进行体外培养,分别加入高、低剂量的健骨二仙丸提取物并与高、低剂量Premarin组、阴性组、Tamoxifen对照组、Tamoxifen阻断对照组进行对照,用Western免疫印迹检测蛋白激酶C、抗凋亡蛋白Bag-1表达。结果:健骨二仙丸提取物能明显提高体外成骨细胞PKC的表达量和抗凋亡蛋白Bag-1的表达量。结论:健骨二仙丸提取物可促进成骨细胞增殖并明显提高PKC和抗凋亡蛋白Bag-1的表达量是其防治骨质疏松的机理之一。 Objective:To explore the effect of Jiangu Erxian Pill Extract(JEPE) on rat osteoblast cells in vitro.Methods:The osteoblasts were separated from the skull of new-born rats,cultured in DMEM containing 10% bovine serum,and then divided into JEPE group(high dosage,low dosage),Premarin group(high dosage,low dosage),blank group,Tamoxifen group,and Tamoxifen inhibiting group.Western blotting was applied to measure the expression of PKC and anti-apoptotic protein BAG-1.Results:The expression of PKC and BAG-1 was significantly improved.Conclusion:JEPE is effective in treating osteoporosis,which might be realized by promoting osteoblast proliferation and enhancing the levels of PKC and BAG-1.
