3-异丁基-1-甲基黄嘌呤对星形胶质细胞环磷酸鸟苷和胶质纤维酸性蛋白的影响 被引量:1

Influence of Phosphodiesterase Inhibitor 3-isobutyl-1-methylxathine on the Cyclic Guanosine Monophosphate and Glial Fibrillary Acidic Protein in Astrocytes

摘要 目的:探讨不同浓度的非特异性磷酸二酯酶(PDEs)抑制剂3-异丁基-1-甲基黄嘌呤(IBMX)对星形胶质细胞中环磷酸鸟苷(cGMP)浓度及胶质纤维酸性蛋白(GFAP)表达的影响。方法:体外纯化培养的SD乳鼠大脑皮质星形胶质细胞经鉴定(GFAP阳性)后随机分成8组,经不同浓度的IBMX干预后,用放射免疫法测定胞内cGMP浓度,免疫荧光测定细胞的GFAP荧光强度值。结果:IBMX 0.3和0.4 mmol·L^(-1)干预后,cGMP浓度及GFAP表达量与对照组相比差异均有统计学意义(P<0.05),两者水平随IBMX干预浓度的增加而增高,呈显著正相关(相关系数为0.95)。结论:IBMX可抑制PDEs的表达和cGMP降解。 Aim: To explore the effects of different concentrations of 3-isobutyl- 1-methylxathine(IBMX) on the concentration of cyclic guanosine monophosphate(cGMP) and expression of glial fibrillary acidic protein (GFAP) in astrocytes. Methods: The incubated and purified (identified by GFAP positive percent) astrocytes taken from cerebral cortex of neonatal SD rat were randomly divided into 8 groups. After the intervention of different concentrations of IBMX, the concentration of cGMP was determined by radioimmunoassay, and the expression of GFAP was measured by immunofluorescence-histochemistry. Results: IBMX could significantly increase the concentration of cGMP and the expression of GFAP, and there was a positive correlation (correlation coefficient is 0.95) between cGMP and GFAP. Conclusion: IBMX might play an important role in decreasing the expression of phosphodiesterase and inhibiting the degradation of cGMP.
