摘要
目的观察高压氧对大鼠脊髓损伤后Fas/Fasl、Caspase-3和凋亡细胞的影响,探讨高压氧治疗脊髓损伤的机制。方法按照Allen’s打击法制备SD大鼠脊髓损伤模型,将20只大鼠损伤后随机分为高压氧组、常氧高压氮组、常压吸氧组和常压空气组,于损伤后6h开始治疗;1、2、3、7和11d取材。采用凋亡细胞检测法检测凋亡细胞,免疫组化法检测Fas/Fasl和Caspase-3表达,光镜下观察并计数分析。结果Fas/Fasl、Caspase-3和凋亡细胞于损伤后1d即升高,Fas/Fasl和Caspase-3于伤后2d即达到高峰,细胞凋亡高峰出现在第3天,高压氧组与各组比较,峰值降低且差异有统计学意义(P〈0.01)。结论高压氧对大鼠继发性脊髓损伤有保护作用,其作用与下调Fas/Fasl的表达,减少Caspase-3,从而抑制细胞凋亡有关。
Objective To investigate effect of hyperbaric oxygen on the expression of Fas/Fasl and apoptosis of neurocytes following spinal cord injury in rats and also to explore the treatment effect and mechanism of HBO on spinal cord injury. Methods The rat model of acute spinal cord injury was developed with Allen's weight dropping. Following spinal cord injury, 120 SD rats were randomly divided into 4 groups: the hyperbaric oxygen (HBO) group, the high pressure nitrogen with normal oxygen group, the normal oxygen group and the normal pressure air group. Segments of the injured spinal cord were collected for morphological studies at 1, 2, 3, 7, 11 days following injury. Apoptosis cells were labeled with Tunel and the expression of caspase-3 and Fas/Fasl was determined with immunohistochemistry. Results were observed under the optical microscope and analyzed with SPSS11.5 statistics software. Results The level of Tunel-positive cells increased at 1 d and peaked at 3 d following injury. The level of Fas/Fasl elevated at 1 d and peaked at 2 d after trauma. Expression of caspase-3 was similar to that of Fas/Fasl. When compared with those of the other groups, Tunel- positive cells in the HBO group decreased with statistical significance at 3 d after injury ( P 〈 0.01 ). Conclusions HBO could decrease the expression of caspase-3 and Fas/Fasl and inhibit cell apoptosis in rats following spinal cord injury, which might play a certain role in the protection of neurons following secondary SCI.
