摘要 目的:了解中国麻风菌株rpoT和SNP基因型特征及地理分布。方法:从32例中国麻风病患者皮损组织中提取DNA。rpoT基因分型采用PCR-直接测序;SNP分型采用PCR-直接测序和PCR-限制性片段长度多态性分析。结果:32份中国菌株的SNP均为3型。32份菌株中,来自我国东南部和藏族地区的12份菌株rpoT基因为4个拷贝;来自云贵川的20份菌株rpoT基因为3个拷贝。结论:32份菌株均为SNP3型,但我国不同地区rpoT基因型不一致,有待进一步研究。 Objective: To understand the characteristics and geographical distribution of the genotypic profiles of M. leprae based on rpoT and SNP in China. Methods: M. leprae DNA was extracted from skin biopsies of thirty - two leprosy patients rpoT genotyping was performed by PCR- sequencing and SNP genotyping by PCR- sequencing and PCR - RFLP. Results: All the thirty - two isolates were SNP type 3 but the allele for rpoT gene was three or four - copy. Of the thirty - two isolates, twelve f?om the southeast region and Tibet were four- copy; twenty isolates from Yunnan, Guizhou and Sichuan three- copy in rpoT gene. Conclusion: The 32 isolates were SNP type 3 but the rpoT allele of M. leprae obtained from different regions is different. The reason is not clear and the distribution of rpoT allele needs to be studied in a large sample.
