摘要
目的:评价2型单纯疱疹病毒(HSV-2)对人永生化角质形成细胞(HaCaT)分泌Ⅰ型干扰素(IFN-α、IFN-β)的影响,探讨角质形成细胞(KC)对HSV-2感染的免疫防御机制。方法:以HSV-2感染HaCaT细胞,采用荧光实时定量PCR检测感染后24 h、48 h、72 hIFN-α1、α2、β1 mRNA的表达。结果:HaCaT细胞静息状态存在IFN-α1、α2、β1三种细胞因子的mRNA表达,但HSV-2感染HaCaT细胞后IFN-α1、α2、β1 mRNA的表达较对照组明显升高(P<0.05),且在72 h内随时间的递增而逐渐升高。结论:在HSV-2感染KC的初期,KC分泌的Ⅰ型干扰素参与HSV-2感染的免疫防御反应,对清除HSV-2及防止其扩散有积极作用。
Objective: To evaluate the effect of HSV- 2 on interferons (IFNs) secreted by human eternal cell line (HaCaT), and to explore the mechanism of immune defensive function of human keratinocyte (KC) against HSV- 2 infection. Methods: HaCaT cells were infected with HSV - 2, and the expression of IFN - a1, α2 and β1 were detected by Real - time PCR at 24 h, 48 h and 72 h after infection. Results: Although expression of IFN - al, a2 and β1 mRNA was detected noninfected HaCaT cells, after HSV- 2 infection, expression was gradually increased and in a time dependent manner within 72 h. Conclusion: The interferons secreted by KCs after initial infection of HSV - 2 is involved in the defence reaction against the infection which may play positive role in prevention from spreading of the infeection.
