摘要
目的建立噪声易感豚鼠模型。方法将103只雄性健康豚鼠暴露于110 dBA窄带噪声3 h。检测噪声暴露前后豚鼠畸变产物耳声发射(distortion product otoacoustic emission,DPOAE)和听性脑干反应(auditory brainstem response,ABR),扫描电镜下观察豚鼠耳蜗形态学改变。利用百分位数法计算豚鼠双耳暂时性阈移均值的90%参考值范围,在此基础上确定易感豚鼠和不易感豚鼠。比较易感组与不易感组豚鼠噪声暴露前后听觉功能和耳蜗形态学改变的差异。结果豚鼠双耳暂时性阈移平均值的90%参考值范围为[18.2 dB,31.7 dB],易感豚鼠7只,不易感豚鼠22只。噪声暴露后,在畸变频率1 409 Hz和2 000 Hz处,易感组DPOAE幅值小于不易感组(P<0.05)。易感组双耳暂时性阈移均值大于不易感组(P<0.05)。与不易感组比较,易感组耳蜗第3回和第4回结构损伤严重。结论成功建立噪声易感豚鼠模型。
Objective To establish the noise-susceptible guinea pig models. Methods One hundred and three male healthy guinea pigs were exposed to 110 dBA narrow-band noise for 3 hours. The distortion product otoacoustic emission (DPOAE) and auditory brainstem response of all guinea pigs were detected before and after the noise exposure. The morphological changes were observed under scanning electron microscope. By using percentile methods ,the 90% reference value of all guinea pigs was determined, through which noise susceptible and unsusceptible guinea pigs were distinguished. Finally, a comparison was made between the two groups for their difference in the auditory function and cochlear morphological structure. Results Percentile algorithm showed that the 90% reference value of guinea pigs' mean value of left and right temporary threshold shift after the noise exposure was within a range of 18.2 dB to 31.7 dB. The number of noise-susceptible and unsusceptible guinea pigs was measured to be 7 and 22 respectively. It could be seen that the DPOAE amplitude of susceptible group was smaller than that of the unsusceptible group at distortion frequency of 1 409 Hz and 2 000 Hz ( P 〈 0.05 ). The mean value of the left and right temporary threshold shift for the susceptible group was greater than that of the unsusceptible group(P 〈 0.05 ). Lesion to the cochlea of the susceptible group was more serious than that of the unsusceptible group,with its chief manifestations in the structure of the third circle and the fourth circle. Conclusions The noise susceptible guinea pig models were established successfully.
