摘要
目的了解三氧化二砷(arsenic trioxide,ATO)对人视网膜母细胞瘤SO—Rb50瘤细胞株成瘤性的影响。方法取出生14d的SD大鼠10只,用含24μmol/l三氧化二砷的5×10^7m1 SO—Rb50瘤细胞悬液0.03ml注射到大鼠任一眼前房;另一眼前房注射同量不含三氧化二砷的瘤细胞悬液作为对照,每日裂隙灯观察前房肿瘤形成的情况;接种后20d,按照接种液是否含有三氧化二砷分为两组,分别挖出荷瘤大鼠眼内的肿瘤组织测定谷胱苷肽过氧化物酶活性;通过统计学分析两组前房肿瘤形成的情况及肿瘤组织谷胱苷肽过氧化物酶活性的差异。结果含24μmol/l三氧化二砷的瘤细胞悬液注射前房后成瘤时间平均为6日(对照组为[日),明显延长(t=7.21,P〈0.01),肿瘤性新生血管形成时间平均为13.8日(对照组为9.6日),也明显延长(t=9.35,P〈0.01);肿瘤组织的谷胱苷肽过氧化物酶活性A340均值为0.144(对照组均值为0.135),活性明显增强(t=9.74,P〈0.01)。结论24μmol/l三氧化二砷能够显著延长SO-Rb50瘤细胞悬液注射前房后成瘤的时间及肿瘤性新生血管形成时间,形成的瘤组织谷胱苷肽过氧化物酶活性明显增强。
Objective To investigate the effect of arsenic trioxide (ATO) on the tumorigenicity of human retinoblastoma cells SO-Rb50. Methods Ten 14-days old SD rats were used in this study. The 0.03ml of 5×105/ml SO-Rb50 cells with 24p mol/l ATO were injected into one anterior chamber of the eye, as the contrast, the opposite eye was injected into the same quantity cells only, then the tumor formation in the anterior chamber of those rats were observed by the slit lamp. After implanted 20 days, the tumor tissues were removed and divided into two groups according to whether containing ATO in the inoculation liquid. The Glutathione peroxidase activity was examined and statistical analysis was performed.Results The tumors and the new vessels of the ones received SO-Rb50 cells with 24p mol/1 ATO injected into anterior chamber had formed in average 6 days and 13.8 days respectively, both of which were longer than that of tumor cells injection only group, which were 2 days ( t=-7.21, P〈0.01 ) and 9.6 days (t=-9.35, P〈0.01 ) respectively. The Glutathione peroxidase activity of the A340 of the tumor tissues was 0.144, which was stronger than the contrast of 0.135 (t=-9.74, P〈0.01 ).Conclusions 24p mol/l ATO can prolonged tumor and tumorous new vessels formation time of SO-Rb50 cells injected into the rats' anterior chambers effectively, and increase the Glutathione peroxidase activity of the tumor tissues.
