苯乙酸联合化疗药物对Hep-2细胞凋亡及Bax表达的影响

摘要 目的初步探讨苯乙酸联合化疗药物促喉癌细胞凋亡的可能机制。方法体外培养Hep-2细胞,将苯乙酸(phenylacetate,PA)、5-氟尿嘧啶(5-Fu)、顺铂(CDDP)及PA分别联合5-Fu、CDDP,5组药物作用于Hep-2细胞24小时,以无用药Hep-2细胞做为对照组,进行吖啶橙染色,分别检测喉癌细胞凋亡百分率,同时通过Bax免疫组织化学染色,观察Hep-2细胞中Bax的表达。结果①单一用药组及联合用药组与对照组相比,细胞凋亡百分率明显高于对照组(P<0.05),联合用药组与单一用药组相比,细胞凋亡百分率高于单一用药组(P<0.05);②单一用药组及联合用药组与对照组相比,Bax蛋白表达水平明显高于对照组(P<0.05),联合用药组与单一用药组相比,Bax蛋白表达水平明显高于单一用药组(P<0.05)。结论①PA联合5-Fu、CDDP对喉癌细胞的作用与单一化疗药物组比,具有明显促进细胞凋亡的作用;②PA联合CDDP、5-Fu抗喉癌细胞的机制与凋亡有关;③PA联合5-Fu、CDDP促进Hep-2细胞凋亡的机制可能与促进促凋亡基因Bax蛋白的表达有关。 OBJECTIVE To study the effects of phenylacetic acid combined with chemotherapeutic drugs(5-FU and CDDP) on apoptosis and Bax protein expression in larynx cancer cell line,Hep-2. METHODS Hep-2 cells were cultured in vitro.After 24 hours of culture,Hep-2 cells were randomly divided into 6 groups:control group,PA,5-Fu,CDDP,PA with 5-Fu and PA with CDDP.Using acridine orange staining method and immunohistochemistry method,the effects of phenylacetic acid combined with 5-Fu and CDDP on the apoptosis and bax expression of the larynx cancer cells were studied.RESULTS Apoptosis rates and expression of Bax protein in single medicine group and combined medicine group were both increased compared with that in control group(P0.05). Apoptosis rate and expression of Bax protein in combined medicine group were increased compared with that in single medicine group(P0.05). CONCLUSION①Combined medicines are more efficient to induce Hep-2 cell apoptosis than single medicine;②The anti-larynx cancer mechanism of the PA combined with CDDP or 5-FU may be correlated with the apoptosis they induced;③The mechanism of enhancing apoptosis of Hep-2 cell by the phenylacetic acid with chemotherapeuatic medicine may have some relationship with the up-regulation of the expression of Bax.
