肺气虚模型大鼠结肠水通道蛋白2表达变化及机制研究 被引量:7

摘要 目的:观察肺气虚模型大鼠结肠组织AQP2表达的变化及肺组织、血浆中ET、IL-1β、TNF-α含量变化。为肺与大肠相表里理论提供依据。方法:大鼠20只随机分为模型组和对照组,应用免疫组化方法观察AQP2蛋白含量的变化,应用放免法观察肺组织、血浆中ET、IL-1β、TNF-α含量变化。结果:肺气虚模型组结肠组织AQP2表达低于正常组(P<0.05);肺组织、血浆中ET、IL-1β、TNF-α含量高于正常组(P<0.05)。结论:肺气虚模型大鼠结肠组织AQP2表达减少,肺组织、血浆中ET、IL-1β、TNF-α含量增加。提示肺主"宣降"功能与大肠濡润功能关系的科学内涵,为中医学肺与大肠相表里基本理论提供实验依据。 Objective:To provide experimental evidence for the tradi-tional Chinese medical theory of "lung being connected with large intestine".The effects of colon aquaporin-2 expression and the change of ET,IL-1β,TNF-αin blood and lung tissues in Lung-Qi deficiency model rats was observed.Methods:Twenty healthy rats were divided into the model group and the control group.The method of Immunohisto-chemistry was used to observe the expression of aquaporin-2 in kidney.Radioimmuno-assay was applied to measure the change of ET,IL-1β,TNF-αin blood and lung tissues.Results:The expression of aquaporin-2 in colon was lower in model group than in the control group.ET,IL-1β,TNF-α concentrations of blood and lung tissue in the model group were markedly higher than those in the control group(P0.05).Conclusion:The expression of aquaporin-2 in colon was lower but ET,IL-1βand TNF-α concentrations of blood and lung tissue was higher in Lung-Qi deficiency model rats.One of the substance basis of "lung being connected with large intestine" may be the change of AQP2 expression in the colon tissues.
