摘要
用纯番茄α-D-半乳糖苷酶,作用于B型红细胞及其影泡和水溶性血型物质,均能使它们的B抗原转化为H抗原专一性,定量地释放出D-半乳糖,表明B型红细胞的血型抗原均分布于细胞膜外表面,且全部能被酶转化,番茄α-D-半乳糖苷酶能将人B型红细胞转换成O型红细胞。探索了酶转化完整B型红细胞的条件,通过对转化前后红细胞渗透脆性、膜乙酰胆碱酯酶活性、高铁血红蛋白浓度、三磷酸腺苷及2,3-二磷酸甘油酸含量的测定,表明转化后的红细胞(称为酶转化O型红细胞,缩写为ECO)的结构和功能保持完整,ECO可继续作临床应用研究。
Treatment of human group B erythrocytes, erythrocyte ghosts and membrane aqueous fraction with highly purified tomato α- D - galactosidase resulted in the disappearance of B antigen activity and enhancement of H antigen activity with concomitant release of galactose.The effect of enzymatically treatment upon red cell structure and function was not obvious. The small hemolyisis and the small change in osmatic fragicity and acetyl cholinesterase indicated the membrane structure of ECO - RBC (enzymatically converted B red cell) to be integrity. Similarly the small amount of methemaglobin and small change of ATP and 2,3 - DPG level also pro -ved the ECO - RBC functional integrity.
