摘要
目的采用HPLC法同时检测博落回不同部位原阿片碱、别隐品碱含量。方法采用Luna-C18色谱柱(150 mm×4.6 mm,5μm),以0.1%磷酸加三乙胺缓冲溶液(pH 3.5)-乙腈(78∶22)为流动相,流速为0.8 mL/min,柱温25℃,检测波长284 nm。结果原阿片碱、别隐品碱含量测定方法的线性关系良好,线性范围和相关系数分别为4.2~52μg/mL(r=0.999 4),6.9~86μg/mL(r=0.999 9);平均回收率分别为99.2%、99.4%;RSD分别为0.9%、1.8%(n=6)。结论该方法操作简便、准确、灵敏度高,适合于博落回中原阿片碱、别隐品碱含量的同时测定,不同部位两种生物碱均存在一定的差异。
Objective To establish a HPLC method for the determination of protopine and allocrytopine contents in Macleaya cordata (willd.)R.Br ,so as to investigate their contents in different parts. Method The analysis was performed on a Luna-C18 column(150 mm×4.6 mm, 5μm)with acetonitrile -0.1% Triethanolamine+Phosphoric Acid as mobile phase. The flow rate was 0.8 mL/min. The column temperature was 25℃ and the detection wavelength was 284 nm. Results A good linearity was obtained with the correlation coefficient ranged from 4.2-52 μg/mL (r=0.999 4), 6.9-86 μg/mL (r=0.999 9). The average recoveries were 99.2%,99.4% with corresponding RSDs of 0.9%,1.8%,respective(n=6). Conclusion The methods is simple and accurate with a good sensitivity. It can be used for determining protopine and allocrytopine in Maeleaya eordata (willd.)R.Br. The contents of protopine and allocrytopine are different in different parts of Macleaya cordata (willd.)R.Br.
