摘要
报道了嗜碱芽孢杆菌NTT33产生的碱性β-甘露聚糖酶经纯化后,在PAGE电泳上呈现一条蛋白带,分子量为3.89×107,等电点为3.0~4.0,酶反应最适pH为9.0~10.0,最适作用温度为80℃,稳定pH为6.0~9.0,稳定温度为60℃以下.金属离子Cu2+,Ba2+,Mg2+,Al3+,Co2+对该酶有一定的激活作用,而Ag+,Hg2+,Mn2+对该酶有明显抑制作用,经薄层层析鉴定,该酶水解槐豆胶后产生葡萄糖、甘露糖以及低聚糖.
The purification and properties of alkaline β mannanase produced by alkalophilic Bacillus sp. NTT33 were reported. Only one kind of alkline β mannanase was obtained . The molecular weight and pI value of the β mannanase were 38 900 and 3.0 ̄4.0 , respectively. The optimal pH and temperiture for β mannanase were 9.0 ̄10.0 and 80℃. The enzyme showed a high stability below 60℃ and at pH 6.0 ̄8.0. The activity of the enzyme was enhanced by Cu 2+ ,Ba 2+ ,Mg 2+ , Al 3+ ,Co 2+ and was inhibited by Ag +,Hg 2+ , Mn 2+ . The major components that the enzyme hydrolysed locust bean gum were glucose, mannose and oligosaccharides.
