摘要
目的探讨傣族和汉族志愿者细胞色素P450 1A2^*1C(CYP1A2^*1C)基因的多态性。方法云南省德宏州117名傣族志愿者和112名汉族志愿者纳入本研究。应用聚合酶链反应-限制性片段长度多态性的方法进行CYP1A2—2964位点分型,检测CYP1A2—2964位点的基因多态性。结果云南省德宏州117名傣族志愿者中,45名为野生型纯合子(G/G);63名为杂合子(G/A);9名为突变型纯合子(A/A);112名汉族志愿者中,63名为野生型纯合子(G/G);44名为杂合子(G/A);5名为突变型纯合子(A/A),傣族与汉族志愿者G-2964A基因型分布差异有统计学意义(P〈0.05);G-2964A基因频率经检验分布符合H—W平衡定律;德宏州傣族和汉族志愿者A-2964位点等位基因频率分别为35%(95%可信区间30%~40%)和24%(95%可信区间20%~30%),傣族和汉族志愿者间差异有统计学意义(P〈0.05);云南省德宏州傣族志愿者A-2964等位基因频率与其他地区人群比较差异有统计学意义(P〈0.05)。结论在云南省德宏州CYP1A2^*1C基因多态性是引起傣族和汉族人药理学特性个体和种族差异的遗传因素之一。
Objective To investigate cytochrome P450 1 A2^* 1 C gene polymorphism in Dai and Han nationality volunteers from Dehong autonomous prefecture in Yunnan province. Methods One hundred and seventeen Dai and 112 Han nationality volunteers from Dehong autonomous prefecture in Yunnan province were enrolled in this study. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was employed in genotyping analysis. Results There were 45 wild-type homozygotes (G/G), 63 heterozygotes (G/A) and 9 homozygotes among 117 Dai nationality volunteers, while 63 wild-type homozygotes (G/G), 44 heterozygotes (G/A) and 5 homozygots among 112 Han nationality volunteers. There was significant difference in the incidences of the genotypes between the two populations ( P 〈 0.05 ). The distribution of the genotypes of either population was in Hardy-Weinberg equilibrium. The frequency of the allele A in the locus-2964 of CYP1A2 was 35% (95% CI 30%-40% ) and 24% (95% CI 20%-30%) respectively in Dai and Han nationality volunteers. There was significant difference in the frequency between two populations ( P 〈 0.05 ). There was also significant difference in the frequency between Dai nationality volunteers and the populations of other regions. Conclusion CYP1 A2^* 1C gene polymorphism is one of factors of producing individual and racial variation in pharmacology in Dai and Han people from Dehong autonomous prefecture in Yunnan province.
