摘要
对链霉菌YH-2进行紫外、微波与硫酸二乙酯诱变,筛选出一株高产突变菌株YH-UV-2,并对其发酵条件进行了优化,结果表明菌株YH-UV-2最适发酵条件为:2%葡萄糖,1%酵母膏,0.05%K2HPO4.3H2O,0.05%NaCl,0.05%MgSO4.7H2O,0.001%FeSO4.7H2O,32℃,pH 7.6,接种量8%,菌株YH-UV-2的抑菌活性较原始菌株提高了30.41%。对菌株YH-UV-2所产抑菌物质的研究结果表明,该抗生素为多酚类化合物,热稳定性良好,pH≤8时稳定;经氯仿-甲醇提取得红褐色粗提物;MIC为68.3μg/mL。
A high-yield strain YH-UV-2 was obtained by ultraviolet light, microwave and DES mutation, and its fermentation medium was optimaized. The results show that the optimal conditions for fermentation of the strain YH-UV-2 were as follows: 2% glucose, 1% yeast extract, 0. 05% Kz/qPO4 · 3HzO, 0. 05% NaCI, 0. 05% MgSO4 ·7HzO and 0. 001% FeSO4 · 7HzO, the culture temperature 32 ℃, pH 7.6, and inoc- ulum 8%. The antimicrobial activity of YH-UV-2 was increased by 30.41% in comparison with the o- riginal strain. Menawhile, the results of the studies of antibacterial substance show that the antibacte- rial substance belonged to polyphenols, and had good thermal stability as the pH value was less than 8. Rufous substance was obtained by chloroform-methanol extraction. The MIC was 68. 3 μg/mL.
