五苓散对水合氯醛介导的大鼠意识障碍影响的实验研究

摘要 目的:观察运用经典方剂五苓散对水合氯醛介导的意识障碍的影响效果并分析其作用机制。方法:①将30只中年Wistar大鼠随机分为造模前6小时灌药组、造模前3小时灌药组、造模前l1小时灌药组各10只,(模型为采用水合氯醛腹腔注射模拟短暂意识障碍状态的Wistar大鼠)。②每组分别于造模前6小时、造模前3小时、造模前1小时给予生理盐水灌服,观察各组大鼠由注射水合氯醛至昏迷的时间以及由昏迷至清醒的时间,经过24小时药物清除后,再分别于造模前6小时、造模前3小时、造模前1小时给予经方五苓散剂灌服(由茯苓、猪苓、白术、泽泻、桂枝组成),并观察各组大鼠由注射至昏迷的时间以及由昏迷至清醒的时间,并对数据进行统计学分析。结果:各组灌服生理盐水与灌服五苓散自身对比显示灌服五苓散的昏迷时间(由腹腔注射至昏迷的时间)均较灌服生理盐水明显延长(P〈0.01),清醒时间均较灌服生理盐水明显缩短(P〈0,01);造模前6小时灌药组的昏迷时间比造模前3小时灌药组显著延长(P〈0.01)且清醒时间比造模前3小时灌药组显著缩短(P〈0.01);造模前3小时灌药组的昏迷时间及清醒时间与造模前1小时灌药组的昏迷时问和清醒时间无显著性差异(P〉0.05)。结论:经方五苓散于适当的时间作用于水合氯醛介导的意识障碍的大鼠模型后,能有效抑制水合氯醛对中枢神经系统的抑制作用,并减短其对中枢神经系统的作用时间,加快其从。净脏排出的速度。 Objective: To observe the use of the classic recipe of Wuling Powder on chloral hydrate-mediated effects of disturbance of consciousness and analyze its mechanism. Methods: ①the 30 middle-aged Wistar rats were randomly divided into six hours before modeling irrigated drug group, 3 hours before modeling irrigated drug group, 1 hour before modeling irrigated treatment group of 10 rats (model for the introduction of chloral aldehyde intraperitoneal injection of simulated short-term disturbance of consciousness state of Wistar rat). ② Each modeling, respectively, the first 6 hours of modeling the first 3 hours and 1 hour before making model for saline irrigation services to observe the rats by injection of chloral hydrate to the unconscious time and from a coma to awake time, After 24 hours of drug removed, re-modeling, respectively, the first 6 hours of modeling the first 3 hours and 1 hour before making model for agent fed through the side of Wuling Powder (from Poria, Polyporus umbellatus, Atractylodes, Alisma, Guizhi composition ), and observed in rats injected into a coma from the time, and awake from the coma to the time, and the data were statistically analyzed. Results: The group fed with saline and fed with Wu Ling San Wu Ling San's own comparison showed coma gavage time (by intraperitoneal injection to the unconscious of the time) was significantly longer than those fed with normal saline (P〈0.01), awake time, was significantly shorter than those fed with normal saline (P〈0.01); made 6 hours before mold filling drug group coma longer than 3 hours before modeling irrigated treatment group was significantly prolonged (P〈0.01) and awake longer than 3 hours before modeling perfusion group were significantly shorter (P〈0.01); modeling 3 hours before perfusion time and awake coma group modeling time 1 hour before perfusion time and awake coma group time there was no significant difference (P〉0.05).Conclusion: The side of Wuling Powder at an appropriate time to act on the chloral hydrate-mediated disturbance of consciousness of the rat model, we could effectively inhibit the chloral hydrate inhibition on the central nervous system, and to shorten their central nervous system, action time, speed up the rate of discharge from the kidneys.
