摘要
建立在麦槽水解液中.单糖、有机酸和糖醛酸等的高效液相色谱测定法.在确定的色谱条件下,葡萄糖、阿拉伯糖、鼠李糖、甲酸、乙酸、葡萄糖醛酸、半乳糖醛酸的分离状况良好,其峰面积与质量浓度的线性关系良好.相关系数联值为0.9912~0.9997.回收率为99.40%~105.90%.木糖、半乳糖、甘露糖在AminexHPX-87H住上不能分离.但可用ZORBAX Carbohydrate Analysis柱分离.采用示差折光检测器检测,柱温为30℃.流动相中乙睛和水的体积比为80:20,流速为1.0mL·min^-1,进样量为0.02mL,外标法定量,结果表明.其峰面积与浓度线性关系良好,相关系数R^2值为0.9959~0.9991,回收率为99.99%~102.70%.
High performance liquid chromatography (HPLC) method was constructed to test individual monosaecharides and organic acids in the liquors of acid hydrolysis on brewery's spent grain. Mixture of the glucose, arabinose, rharnnose, formic acid, acetic acid, glueuronic acid and galacturonic acid was measured in the given conditions. The correlation coefficient of these matters was 0. 991 2 to 0. 999 7 and the corresponding average recovery was 99. 40% to 105.90%. For the case where, xylose, galactose and mannose can not be separated by Aminex HPX-87H column, ZORBAX Carbohydrate Analysis column was used for further separation. The operating conditions were as follows.. ZORBAX Carbohydrate Analysis column with differential refractive index detector, 30℃, elution influent acetonitrile : water (80: 20, volume ratio), flow rate 1.0 mL · min^-1 and injection volume 0. 02 mL. Under these conditions, the correlation coefficient was 0. 995 9 to 0. 999 1 and the corresponding average recovery was 99. 99% to 102. 70%.
