摘要
目的研究拳参正丁醇提取物(PBNA)的水溶性成分PBNA-413对离体心脏缺血再灌注损伤时的抗氧化作用。方法应用大鼠离体心肌缺血再灌注损伤模型,在灌流液中加入高、中、低剂量的拳参正丁醇提取液进行再灌注。再灌注结束后,测定心肌组织乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD),谷胱甘肽过氧化物酶(GSH-Px)的活性及丙二醛(MDA)的含量。结果各剂量的PBNA-413治疗后,均能提高心肌组织LDH(P<0.01)、SOD(P<0.05)、GSH-Px(P<0.01)的活性,升高NO(P<0.05)含量,降低MDA(P<0.001)及LD(P<0.01)的含量,且呈明显的剂量依赖性。结论拳参正丁醇提取物可通过提高抗氧化酶系统的功能、减少心肌酶的释放而保护缺血再灌注心肌。
Objective To explore the anti -oxidative effects of water -soluble constituent ofpolygonum bistorta L. n -butyl alco-hol (PBNA -413 ) in isolated myocardial ischemia/reperfusion injury. Methods A rat myocardial ischemia/reperfusion injury was made and treated with PBNA -413 (high dose, medium dose and low dose). The content of malondiahtehyde(MDA) and the activities of lactate dehydrogenase (LDH) , superoxide dismutase (SOD) and glutathion peroxidase GSH- Px) were investigated. Results PBNA- 413 coud significantly increase the enzymatic activities of LDH, SOD, GSH- Px, elevate the concentration of NO, and decrease the levels of MDA and LD in myocardial tissue. Conclusion PBNA -413 has a therapeutic effect on myocardial ischemia/reperfusion injury through its anti -oxidation in myocardial tissue
