摘要
目的研究激素+滋阴清热方治疗前、中、后阴虚内热证红斑狼疮(SLE)外周血单一核细胞(PBMC)凋亡相关BCL-2基因家族的动态表达。方法阴虚内热证SLE患者治疗前、中、后三次及健康对照组外周血单一核细胞基因表达谱芯片检测,对BCL-2基因家族成员数据进行分析。结果找到BCL-2基因家族基因16个,其中BCL-2、BAG3治疗前、中、后均明显下调,而BAG1、BCL6、BCLA21治疗后表达均上调。结论阴虚内热证SLE患者PBMC存在BCL-2基因家族凋亡基因的异常,激素与滋阴清热方治疗后抑制/促进凋亡的BCL-2基因比例发生明显改变,细胞凋亡减少。BCL6可能是中医药等治疗SLE的靶基因之一。提示研究不同证型SLE治疗前后T、B淋巴细胞亚型的凋亡情况及凋亡机制,是合理评价凋亡在SLE发病及诊治中的作用的关键。凋亡基因的调控可能是滋阴清热方实现阴平阳秘的内在机制之一,有待深入研究。
Objective To study the dynamic expression of the apoptosis - associated BCL - 2 gene family in the peripheral blood mononnclear cells (PBMC) of the heat deficiency patients with lupus erythematosus (SLE) before, during and after treatment with hormone and Ziyin Qingre formula. Methods Before, during and after treatment, peripheral blood mononuclear cells of the SLE patients with "Yin" asthenia generating intrinsic heat were separated, then cDNA microarray chip was used to analyze and compard with a healthy control group,in this part, the data of BCL -2 gene family members were selected out and analyzed. Restilts 16 BCL - 2 family gene were found. The expression of BCL - 2 and BAG3 were significantly reduced before, during and after treatment, while the expression of BAG1, BCL6 and BCLA21 was increased after treatment. Conclusion Abnormalities of BCL - 2 family gene in PBMC of SLE patients with "Yin" deficiency heat were observed. Apoptosis suppression and promotion gene of BCL -2 family were changed by the hormones and Ziyin Qingre formula treatment which led to reducing apoptosis. BCL6 may be one of the target gene which was affected by the treatment of Chinese medicine. To study the different subtypes apoptosis and apoptosis mechanisms of T and B lymphocyte of SLE with different syndrome pattern before and after treatment is very important for estimation on the role of apoptosis in SLE pathogenesis, diagnosis and treatment. Regulation of apoptotic genes may be one of the mechanisms of Ziyin Qingre formula to achieve " Yin" and " Yang" in a relative equilibrium.
