肺腺癌A549细胞STAT3基因沉默对细胞周期分布和凋亡水平变化的影响

摘要 目的:研究沉默STAT3基因表达前后肺腺癌A549细胞的细胞周期分布状况和细胞凋亡水平变化及其意义。方法:利用流式细胞仪(FCM)检测RNA干扰(RNAi)技术沉默STAT3基因表达前后肺腺癌A549细胞的细胞周期状况和细胞凋亡水平变化。结果:STAT3沉默后,肺腺癌A549细胞大量处于G0/G1期(P<0.05),而S、G2/M期的细胞相对减少(P<0.05);并且细胞凋亡明显增多,前48h增长明显(P<0.05),48h后增长趋于平稳(P>0.05),但仍能较长时间维持较高水平。结论:STAT3siRNA能阻断A549细胞的细胞周期,使细胞周期停留在G0/G1期,不能进入S期,不能完成DNA的合成,无法进入M期,未完成细胞的分裂,并能诱导肺腺癌A549细胞的凋亡,提示STAT3的表达与A549细胞的细胞周期和凋亡密切相关。 Objective: To investigate the changes of cell cycle distribution and apoptosis in lung adenocarcinoma A549 cells before and after the STAT3 gene silencing and its significance. Methods: Before and after RNA interference (RNAi) technology silencing STAT3 gene expression in lung adenocarcinoma A549 ceils ,the changes the cell cycle status and the level of apoptosis were detected by flow cytometry. Results:After STAT3 was silent, the lung adenocarcinoma A549 cells in the G0/G1 phase increased (P〈0.05), while in the S, G2/M phase decreased (P〈0.05); and apoptosis increased;in the first 48h ,the growth was significant (P〈0.05), after 48h ,the growth stabilized (P〉0.05), but still maintain a high level for a long time. Conclusion: STAT3siRNA can block the cell cycle of A549 ceils and induce the apoptosis.It suggests that STAT3 expression in A549 cells are closely related with the cell cycle and apoptosis.
