摘要
[目的]研究猪初情期NPY-Y1 mRNA在下丘脑、垂体和卵巢的分布定位。[方法]选取60日龄、体重为20 kg和160日龄、体重为80 kg的苏姜母猪各3头,麻醉后,取出其下丘脑、垂体和卵巢,制成冰冻切片。采用原位杂交技术检测各样品中NPY-Y1 mRNA的表达定位,并用PBS液取代杂交液作阴性对照。[结果]不同发育阶段的下丘脑、垂体和卵巢中均检测到NPY-Y1 mRNA的阳性杂交信号,且初情期前期(60日龄)样品的阳性信号较初情期(160日龄)强。[结论]下丘脑-垂体-卵巢轴内分布有NPY-Y1 mRNA,其对雌性生殖过程具有调控作用。
[Objective]To study the disposition and localization of NPY-Y1 mRNA at hypothalamus,pituitary and ovary in puberty.[Method] After three 60 day-old Su Jiang sows weighing 20 kg and three 160 day-old Su Jiang sows weighing 80 kg were selected and anaesthetized, and the hypothalamus,pituitary and ovary were taken out for preparation of frozen sections.The expression and localization of NPY-Y1 mRNA in hypothalamus-pituitary-ovary Axis was performed by in situ hybridization(ISH).The phosphate buffer solution(PBS) instead of hybridization solution was used as negative control.[Result]The positive hybridization signals of NPY-Y1 mRNA were detected in hypothalamus,pituitary, and ovaries at different developmental stages.Moreover,the samples positive signal of 60 day-old sows were stronger than those of 60 day-old sows.[Conclusion]NPY-Y1 mRNA distribute in the hypothalamus-pituitary-ovarian axis,and has regulatory effect on female reproductive process.
