摘要 目的:研究E6相关蛋白与前列腺癌细胞迁移浸润的相关性,为探讨前列腺癌的浸润转移机制提供实验依据。方法:应用电穿孔方法将线性化的PrevTet-off-in调控质粒和PrevTRE2-flag-E6AP重组质粒导入前列腺癌细胞(Lncap),建立稳定表达E6-AP的前列腺癌细胞。应用MTT实验检测细胞的增殖;应用Matrigel Invasion Chamber检测肿瘤细胞迁移浸润特性。结果:MTT结果显示,稳定转染表达E6-AP的前列腺癌细胞比未转染的细胞具有较强的增殖活性;在细胞的迁移浸润试验中,表达E6-AP的稳定转染细胞具有较强的爬行与附着能力。结论:E6相关蛋白与前列腺癌细胞迁移浸润性密切相关。 Objeetive: The carcinoma of prostate is a prostate gland familiar malignant tumor. It transfers gradually People pay attention to its migration and infiltration . This text mainly studies the relativity that the E6 - AP with carcinoma of prostate cell migration and infiltration . For inquirying into the gradually transfer of the carcinoma of prostate mechanism to provide an experiment. Methods: Apply the electricity boring method turns line of PrevTet - off- in,adjusting to control the quality grain and the PrevTRE2 - flag - E6 AP reorganization plasmid dueting prostate gland cancer cell( Lneap), and build up the prostate gland eancer of the stability expression E6 - AP. Apply the M TT experiment an examination cell to propagate. The applied Matrigel Invasion Chamber examination tumor cell moves gradually characteristic. Results: The MTT shows the stability turns the cell that the prostate gland cancer cell of dye the expression E6 - AP ratio didn't turn to dye to have as a result stronger propagate an activity. The stability that expresses E6 - AP turns to dye cell to have stronger crawl along and adhere to an ability. Conclusion: The E6 - AP albumen and carcinoma of prostate cell migration and infiltration is closely related
