海南地区黎族人类血小板抗原1～17系统基因多态性分析

Analysis of gene polymorphism human platelet antigen 1～17 system in Li ethnic group in Hainan.

目的研究海南地区黎族人群血小板特异性抗原1~17系统基因多态性区域分布特点,建立HPA基因型资料库。方法采用PCR-SSP技术对180名黎族人进行了HPA1~17系统基因分型,计算基因型频率、基因频率。结果黎族人HPA的基因频率分别为:HPA-2a0.9972、-2b0.0028、-3a0.4889、-3b0.5111、-5a0.9667、-5b0.0333、-6a0.9972、-6b0.0028、-15a0.4250、-15b0.5750;HPA-3、15系统表现出较高的杂合度,其中3a/3a、3a/3b、3b/3b基因型频率分别为0.2611、0.4556、0.2834;15a/15a、15a/15b、15b/15b基因型频率分别为0.2167、0.4167、0.3667;HPA-1、-4、-7^-14、-16、-17系统只检出a基因,呈单特异性。结论海南地区黎族人HPA-3、15系统具有多态性,是HPA配合性输注关注重点。在随机血小板输注中,供受者HPA-3系统不配合的机会为37.49%、HPA-15系统不配合的机会为36.93%。黎族人HPA-17系统均为a/a基因型。

Objective To analyze the regional distribution feature of gene polymorphism of human platelet antigen 1 - 17 system in Li ethnic group in Hainan Province for preparation of setting up HPA genotype data bank. Methods HPA1-17 system genotyping of 180 Li ethnic group was conducted by using PCR-SSP and the genotype frequency and gene frequency were calculated. Results The gene frequency of Li ethnic group were HPA-2a 0.9972, -2b 0.0028, -3a 0.4889,-3b 0.5111,-5a 0.9667,-5b 0.0333,-6a 0.9972,-6b 0.0028,-15a 0.4250 and -15b 0.5750;high heterozygosity of HPA-3 and 15 systems were observed. The genotype frequency of 3a/3a ,3a/3b and 3b/3b were 0.2611,0.4556,0.2834,15a/ 15a,15a/15b and 15b/15b; while the gene frequency were 0.2167,0.4167 and 0.3667; Only monospecific gene a was observed in the systems of HPA-1,-4,-7--14,-16 and -17. Conclusion The system of HPA-3 and 15 in Li ethnic group in Hainan are polymorphic and it is the key point of HPA suitable infusion. The incompatibility rates of donorrecipient HPA-3 and HPA-15 were 37.49% and 36.93% in random HPA infusion.All the genotype of system of HPA-17 in Li ethnic group in Hainan is a/a .
