宁夏地区黄牛博尔纳病病毒自然感染状况的探讨 被引量:3

摘要 目的了解宁夏地区黄牛博尔纳病病毒(Borna disease virus,BDV)的自然感染情况,对检测到的BDVp24基因阳性扩增片段进行核苷酸序列比对,探讨其与国际标准病毒株及其分离株的同源性。方法采用荧光定量巢式逆转录酶聚合酶链反应(FQ-nRT-PCR)检测宁夏地区205例健康牛外周血单个核细胞(PB-MCs)BDVp24基因片段,并对其中的阳性扩增产物进行基因序列测定,然后应用BLAST软件以及DNAsist5.0软件对测序结果分析。结果205例中3例阳性,阳性率为1.5%;该BDVp24片段的核苷酸序列与StrainH3915同源性最高达97.67%,与标准病毒株H1766同源性为96.51%,与StrainV/FR的同源性为95.35%,且它们编码的氨基酸相同。结论宁夏地区黄牛中存在动物源性BDV的自然感染,该BDVp24核苷酸序列与标准病毒株具有高度同源性。 Objective To investigate the natural infection of BDV in cows of Ningxia and to explore if the nucleotide sequence in BDV p24 was homophylic with the overseas strains detected from human and animals. Methods The p24 fragment of BDV was detected by nested reverse transcriptase polymerase chain reaction with Fluorescence Quantitative PCR(FQ-nRT-PCR) in PBMCs from 205 cows. The products of FQ-nRT-PCR from positive samples were cloned and sequenced before compared with those of the overseas strains by BLAST and DNAsist 5.0. Results There were 3 positive( 1.5 % ). BDV p24 in cows from Ningxia was homophylic with strain V/FR(95.35% ), strain H1766(96.51% ) and strain H3915(97.67% ). However, their amino acid sequence remained the same. Conclusion There is BDV natural infection probably originated from cows of Ningxia. The gene sequence is high homophylie with that of standard strain.
