首乌补肾酒中何首乌的含量测定方法研究

Study on Content Determination of Radix Polygoni Multiflori in Shouwu Bushenjiu by HPLC

目的建立首乌补肾酒中何首乌的含量测定方法。方法采用Agilent 1100高效液相色谱仪,Thermo Hypersil ODS-2(250 mm×4.6 mm,5μm,柱号1254013M),乙腈-水(15∶85)为流动相,检测波长为320 nm。结果使用高效液相色谱法可以准确有效地测定首乌补肾酒中2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量。结论该方法在11.08～110.80μg/ml的浓度范围内线性关系良好(r=0.9998,n=5),平均回收率为100.90%,相对标准偏差(RSD)=0.5%(n=6),方法准确,适用于首乌补肾酒中何首乌的含量测定。

Objective To establish an HPLC method for the content determination of radix polygoni muhiflori in Shouwu Bushenjiu. Methods The determination was performed on a Thermo Hypersil ODS-2 （250 mm ×4.6 mm,5 μm）. The mobile phase was acetonitrile - water （ 15：85 ）. The detection wavelength was set at 320 nm. Results The content of 2,3,5,4＂- tetera - hydroxystilbeue - 2 - O - β - D - glucoside in Shouwu Bushenjiu could be accurately determined by HPLC. The assay was linear in the range of 11.08 -110.80 μg/ml （r = 0.9998, n = 5 ）. The average recovery was 100. 90%, RSD = 0.5% （ n = 6 ）. Conclusion The method is accurate, and is suitable for the determination of Shouwu Bushenjiu.
