摘要
建立测定不同品种梨中主要有机酸类物质的高效液相色谱法,在以C18反相色谱柱、紫外检测器(波长220nm),甲醇和磷酸盐缓冲液(pH=3.0)作为流动相梯度洗脱,流速为0.6mL/min,柱温为35℃的色谱条件下,一次同时分析梨中7种有机酸仅需5.5min,有机酸标准曲线相关系数均在0.9997以上,回收率在82.9%~91.2%之间。
A quantitative high performance liquid chromatography method with UV detector was developed for the determination of seven organic acids in the pears. A C18 reversed-phase column was applied with methanol- phosphate buffer solution (pH=3.0) as the mobile phase at a flow rate of 0.6 mL/min. The column temperature was 35 ℃ and the UV detection wavelength was 220 nm. The retention times for all of the seven organic acids in the pears were less than 5.5 min. The calibration curves for these organic acids were in good linearity with all of their correlation coefficients were over 0.999 7. Recoveries of these organic acids were in the range 82.9 %-91.2 %.
