转染内皮抑素质粒对人脑胶质肉瘤C6细胞株放射治疗影响的研究

摘要 目的探讨ES对人胶质肉瘤细胞株C6放射治疗的影响。方法通过质粒转染的方法,运用MTT法和流式细胞仪检测了ES以及ES和放射治疗联合作用对C6细胞株增殖和凋亡的影响。结果MTT法检测表明放射治疗对转染pcDNA3.1 ES的C6细胞的生长抑制作用明显增强,细胞增殖明显减慢,差异具有显著性(P<0.05);流式细胞仪检测表明,转染pcDNA3.1 ES后,G0/G1期细胞明显增加(P<0.05),S期细胞明显减少(P<0.01),凋亡率明显上升(P<0.01))。结论ES联合放射治疗可明显抑制人脑胶质肉瘤C6细胞株的生长,二者联用可以产生协同抗瘤作用。 Objective To investigate the co-effect of combination of endostatin (ES) and radiotherapy on the proliferation and apoptosis in human glioma sarcomatosum cell line (C6). Methods The pcDNA 3.1 ES was transfected into C6 cell, and the effect on proliferation and apoprosis in C6 cell was detecled by MTT and flow cytometer. Results The inhibition of radiotherapy to the growth of the C6 cell transfected pcDNA 3.1 ES was enhanced significantly, and the proliferation was slowed significantly by MTr( P 〈 0.05). The flow cytometer detection indicated that after the transfection of pcDNA 3.1 ES, the G0/G1 stage cell rate of C6 cell line was increased significantly( P 〈 0.05 ) , and the decrease in the S stage cell rate and the upgrade of apoptosis were significant (P 〈0.01 ). Conclusion The combination of ES and radiotherapy inhibits the growth of human glioma sarcomatosum cell line (Ca). Their co-use may generate cooperational anti-tumor effect.
