摘要
目的探讨Notch信号通路在宫颈癌发病机制中的作用。方法采用实时荧光定量PCR和流式细胞术,分别从基因转录和蛋白表达水平检测30例宫颈癌患者和30例健康对照者的外周血单个核细胞(PBMCs)中Notch1,Notch2,Notch3,Notch4,以及Notch的配体Jag-ged 1,Jagged 2,Delta 1,Delta 3,Delta 4的表达。结果宫颈癌患者PBMCs中的Notch 1,Notch 2,Jagged1,Jagged 2,Delta 1,Delta 4的mRNA和蛋白的表达均明显高于健康对照组(P<0.01),Notch 3,Notch 4,Delta 3的基因和蛋白的表达在两者中差异均无统计学意义(P>0.05)。结论Notch信号通路的表达在宫颈癌的发病过程中存在一定的变化,可能参与宫颈癌的发生发展,从而为宫颈癌的早期诊断和临床治疗提供新线索。
Objective To investigate the expression changes of notch signal pathway on peripheral blood mononuelear cells (PBMCs) in cervical cancer patients and study the relationship between notch signal pathway and cervical cancer. Methods Real-time PCR and flow cytometry were used to measure the gene transcription and the protein expression levels of receptor Notehl, Notch 2, Notch 3 and Notch 4, and ligand Jagged 1, Jagged 2, Delta 1, Delta 3 and Delta 4in the PBMCs of 30 patients with cervical cancer and 30 normal controls. Results Both the mRNA and protein expression levels of notch 1, notch 2, jagged 1, jagged 2, Delta 1 and Delta 4 were significantly higher in the cervical cancer patients than in the normal controls. There were no differences in the expression levels of notch 3, notch 4 and Delta 3 between the two groups. Conclusion The results indicate the changed expression patterns of Notch receptors and their ligands in cervical cancer, which are informative for further study on the pathogenic mechanism of the disease.
