摘要
目的:建立以蒸发光散射检测法测定山茱萸中熊果酸含量的高效液相分析方法。方法:采用SHMADZU VP-ODS(4.6mm×250.0mm,5μm)色谱柱,流动相为甲醇:0.1%冰醋酸溶液(80:20),流速1.0ml/min,柱温为30℃;Alltech蒸发光散射检测器,载气为氮气,流速:2.0L/min,漂移管温度120℃。结果:熊果酸进样量在0.1048~1.0480μg范围内呈良好的线形关系(r=0.9996),平均回收率为99.4%,RSD值为0.45%。结论:本方法快速简便,结果准确、可靠。
Objective: To determine the content of Ursolic acid in Comus officinalis sieb.et Zucc by HPLC-ELSD. Methods: Ursolic acid was successfully separated on a SHMADZU VP-ODS (4.6 mm×250.0 mm,5 μm) at the ambient temperature, using methanol: 0.1% acetic solution (80:20) as mobile phase. The flow rate was 1.0 ml/min. The drifttube tempera- ture and nitrogen carrier gas flow rate of the ELSD were set at 120℃and 2.0 L/min respectively. Results:The calibration curves were linear in the range of 0.104 8-1.048 0μg for Ursolic acid (r=0.999 6),and the average recovery was 99.4%,RSD was 0.45%. Conclusion:The results of the experiments showed that this method is accurate, rapid, simple and reproducible.
