血管紧张素Ⅱ对肾小管上皮细胞分泌纤连蛋白的影响

摘要 目的:观察血管紧张素Ⅱ(AngⅡ)对体外培养的肾小管上皮细胞分泌细胞外基质成分纤连蛋白的影响。方法:采用免疫印迹法(Westernblot)及细胞免疫荧光法检测10-6mol/LAngⅡ分别刺激肾小管上皮细胞6、12、24、48、72、96h,以及不同浓度(10-6、10-7、10-8mol/L)AngⅡ刺激96h时细胞纤连蛋白的表达。结果:10-6mol/LAngⅡ刺激体外培养的肾小管上皮细胞12h后纤连蛋白表达开始增加,72h达高峰。不同浓度的AngⅡ刺激96h后,纤连蛋白的表达均有增加,10-6mol/L增加最明显,10-7、10-8mol/L次之。结论:AngⅡ能使体外培养的肾小管上皮细胞分泌纤连蛋白增加,并具有时间和浓度依赖性。 Objective:To investigate the effect of angiotensinⅡ(Ang Ⅱ ) on fibronectin(FN) biosynthesis on human renal tubular epithelial cells. Methods:Cultured human renal tubular epithelial cells were treated with Ang Ⅱ at the concentrations of 10^-6 mol/L for 6,12,24,48,72 and 96 hours ; and the cells were treated with Ang 11 at the concentrations of 10^-8, 10 ^-7 and 10^ -6 mol/L for 96 hours. The FN expression in renal tubular cells was determined by both Western blot and immunofluorescence. Results:Fibronectin synthesis was increased by Ang Ⅱ at a concentration of 10^- 6 mol/L in the renal tubular epithelial cells. The expression of FN started to increase after 12 hours, and reached the peak after 72 hours. The synthesis of FN was the most obvious at a concentration of 10 ^- 6 mol/L, and the following was at 10^7and 10^-8 mol/L. Conclusions:It suggests that Ang Ⅱ stimulates FN secretion of human renal tubular epithelial cells in a dose and time-dependent manner.
