Effect of Rho-kinase inhibitor on experimental allergic neuritis

摘要 目的观察ROK抑制剂对EAN发病的影响。方法54只Lewis大鼠随机分为EAN模型组、EAN+ROK抑制剂干预组和完全弗氏佐剂对照组(CFA)组。观察各组大鼠体重变化、发病率、开始发病时间、最高临床评分和组织病理学改变。结果EAN组大鼠在抗原免疫后第17天达到发病高峰;EAN+ROK抑制剂组大鼠与EAN组相比,有体重增加、发病率降低、开始发病时间延迟的趋势,其平均最高临床评分明显降低(P〈0.01),坐骨神经炎性细胞浸润和脱髓鞘减轻,CFA组大鼠无症状。结论ROK抑制剂可以减轻EAN发病程度,抑制Rho/ROK信号通路可能是其作用机制之一。 Objective To study the effect of Rho-kinase inhibitor on experimental allergic neuritis. Methods 54 female Lewis rats were divided into three groups: EAN group, EAN + Rho-kinase inhibitor group, and CFA group. The rats were sacrificed on the 9th day, 17th day, and 26th day after immunized. The changes of weight, EAN incidence, and mean day of onset, mean maximum clinical score, and histopathology were observed. Results The clinical course in EAN group reached peak on the 17th day. Compared with EAN group, the weights of Rho - kinase inhibitor group were increased, while EAN incidence, mean day of onset delay, and the clinical scores in Rho-kinasc inhibitor group were significantly decreased, ( P 〈 0. 01 ) , and the demyelization and inflammation cells infiltrating was ameliorated in spinal nerve. CFA group didnt show any clinical manifestation. Conclusions Rho - kinase inhibitor may ameliorate the development of EAN through inhabiting the Rho/ROK signal pathway.
