摘要
利用SSR标记对24份“禾”品种进行遗传多样性分析,初步建立了其DNA指纹图谱。从145对SSR引物中筛选出21对多态性引物组合,对所有供试材料进行分析,共检测出73个等位基因,平均为3.4762;平均PIC指数为0.4084,范围在0.1175(RM147)-0.6810(RM336)。应用6对引物(RM20A、RM161、RM253、RM280、RM336和RM495)的组合获得了每个品种的SSR特征带,可以将所有供试材料区分开来,从而建立24份“禾”品种的SSR指纹图谱。UPGMA聚类分析表明,在相似系数为0.652处,可将所有材料分为5类,聚类结果表明品种间的亲缘关系与地理来源关系不大。
The DNA fingerprint map of 24 local rice varieties was established from analyzing the genetic diversity by using SSR marker in Guizhou. 73 alleles were detected from tested rice varieties by using 21 pairs of primer combinations with polymorphism screened from 145 pairs of SSR primers. The average number of alleles per locus was 3.4762 and its average PIC index with 0. 4084 varied from 0. 1175 (RM147) -0. 6810 (RM336). The SSR fingerprint maps of 24 local rice varieties were established according to the SSR characteristic bands identified by using six pairs of primer combinations ( RM20A, RM161, RM253, RM280, RM336 and RM495 ). The UPGMA cluster analysis showed that 24 local rice varieties could be divided into five groups at 0.652 of the similar ccefl3cient. The cluster results showed that there was no closer genetic relationship in blood relationship and geographical source among 24 local rice varieties in Guizhou.
