摘要 目的建立前列腺癌细胞PC-3骨转移动物模型,观察肿瘤在骨组织的局部生长及对骨组织的破坏情况,同时观察肿瘤远处器官转移情况。方法16只8周龄BALB/C裸鼠,分为实验组、对照组各8只。BALB/C裸鼠麻醉后,实验组裸鼠将10μl约2×105个PC-3细胞悬液从左侧胫骨近端关节面注入胫骨骨髓腔内,建立骨转移模型。对照组则注入10μl生理盐水。观察裸鼠的活动情况。6周后麻醉裸鼠行X线检查,观察骨破坏情况。然后处死裸鼠,取左侧下肢、肺和肝脏,经固定、石蜡包埋、切片及HE染色后在显微镜下观察组织学情况。结果实验组8只裸鼠骨转移模型建立均成功,其中7只X线检查表现出明显溶骨性破坏。解剖后发现其中6只在骨外也有明显的肿瘤组织生长。显微镜下观察8只裸鼠左侧胫骨内均有肿瘤生长,其中3只发生肺转移。对照组则均未发现明显骨质破坏。结论成功建立了前列腺癌细胞PC-3骨转移模型,模拟了前列腺癌对骨组织的侵袭和破坏及远处转移,为进一步研究前列腺癌对骨破坏的机制及治疗提供了基础。 Objective To establish bone metastasis model of prostate cancer cell line PC-3 for observing local growth and distant metastasis of the tumor and destruction of bone by the tumor. Methods Total 16 male nude mice of 8 weeks old were devided into two groups: experimental group and control group, 8 each group. For 8 mice in the experimental group, 10ul (2× 10^5 )of sus pension of PC-3 cells were injected into left tibia of the mice. And for the control group, the same volume of normal saline were injected. Observe the activities of the mice. 6 weeks after the injection, mice were anesthetized and a plain film was taken to evaluate bone destruction of the tibia. The lungs, livers, and left hind legs were harvested after sacrifice. The specimens were fixed in 10% formalin, embedded in paraffin, stained by H&E and observed microscopically. Results In the experimental group, tumor cells were found in all the tibia of the mice. Tumor growth out of the tibia were found in 6 mice. Lung metastasis were found in 3 mice. Bone destruction were found in 7 mice on X ray film. While in the control group, no tumor and no bone destruction were found. Conclusions Bone metastasis model of prostate cancer cell line PC-3 were successfully established. Tumor invasion, distant metastasis and bone destruction were found in the model. It is a useful tool for in vestigating mechanism of bone destruction and treatment of bone metastasis.
