摘要
目的建立兔眼外源性烟曲霉性眼内炎模型。方法选取健康成年新西兰大白兔24只,随机分为8组进行预实验。分别于右眼玻璃体内接种浓度为(1.0~1.5)×104CFU.L-1、(1.0~1.5)×105CFU.L-1、(1.0~1.5)×106CFU.L-1、(1.0~1.5)×107CFU.L-1、(1.0~1.5)×108CFU.L-1、(1.0~1.5)×109CFU.L-1、(1.0~1.5)×1010CFU.L-1的烟曲霉孢子悬液及生理盐水0.1mL。观察15d后,对眼球进行组织病理学观察,并取玻璃体液进行真菌培养、真菌形态学及分子生物学鉴定。选取能引起典型临床特征的较低浓度孢子悬液进行重复实验。结果玻璃体内接种浓度为(1.0~1.5)×104CFU.L-1、(1.0~1.5)×105CFU.L-1和(1.0~1.5)×106CFU.L-1的新西兰大白兔眼部组织感染体征较轻,3个浓度组成模率为11%;其余浓度组眼部体征较重,成模率为100%。经重复实验确定(1.0~1.5)×107CFU.L-1的烟曲霉孢子悬液可引起新西兰大白兔眼部组织典型的感染体征,眼部组织培养真菌呈阳性,主要内脏和血液培养阴性。结论玻璃体内接种(1.0~1.5)×107CFU.L-1浓度的烟曲霉孢子悬液能够成功建立兔眼外源性烟曲霉性眼内炎模型。
Objective To establish the exogenous aspergillus fumigatus endophthalmitis model in rabbits.Methods Twenty-four healthy adult New-Zealand rabbits were randomly divided into eight groups and the preliminary experiment was carried.The vitreous cavity in right eyes of rabbits in eight groups were injected with (1.0-1.5)×10^4 CFU·L^-1,(1.0-1.5)×10^5 CFU·L^-1,(1.0-1.5)×10^6 CFU·L^-1,(1.0-1.5)×10^7 CFU·L^-1,(1.0-1.5)×10^8 CFU·L^-1,(1.0-1.5)×10^9 CFU·L^-1,(1.0-1.5)×10^10 CFU·L^-1 aspergillus fumigatus suspension and normal saline respectively,0.1 mL in each group.After observed for 15 days,the eyeballs were examined by histopathology,the vitreous liquid were executed to make fungi culture,and then the cultured fungi were identified by morphology and molecular biology.Repeated experiment was taken in the lower concentrations in which typical clinical features could be observed.Results The infective signs in eyes' tissue of the rabbits injected with(1.0-1.5)×10^4 CFU·L^-1,(1.0-1.5)×10^5 CFU·L^-1,(1.0-1.5)×10^6 CFU·L^-1 aspergillus fumigatus suspension were mild,and the rate of successful endophthalmitis model establishment was 11%.The infective signs of other concentrations were serious and all of the rabbits established the endophthalmitis model successfully.Typical fungal infectious signs of eyes' tissue were observed through the repeated experiment in the group injected with(1.0-1.5)×10^7 CFU·L^-1 aspergillus fumigatus suspension,the fungal culture of eyes' tissue was positive,but main organs and blood infection were negative.Conclusion Aspergillus fumigatus suspension with concentration of (1.0-1.5)×10^7 CFU·L^-1 injecting into the vitreous cavity can successfully establish the exogenous aspergillus fumigatus endophthalmitis model in rabbit.
