摘要
目的检测癌-睾丸抗原MAGE-A1在视网膜母细胞瘤(retinoblastoma,RB)组织中的表达情况,探讨其成为RB特异性免疫治疗靶抗原的可能性。方法采用RT-PCR技术研究人RB组织、非肿瘤病变视网膜组织及眼部正常组织中MAGE-A1基因的表达情况;采用免疫组织化学技术对RB组织和正常视网膜组织中MAGE-A1蛋白的表达进行分析。结果MAGE-A1基因在RB组织中的表达率为33.33%;在所有非肿瘤病变视网膜组织及眼部正常组织中则无表达;MAGE-A1基因的表达与患者的各临床指标间无显著相关性(P>0.05)。MAGE-A1蛋白在RB组织中的表达率为20%,在正常视网膜组织中则不表达;MAGE-A1蛋白的表达率与患者年龄相关,年龄≤2岁的患者有表达增高的倾向(P<0.05);眼内期有表达增高的倾向(P<0.01);MAGE-A1蛋白的表达与RB患者的性别、肿瘤的分化程度和大小无显著相关性(P>0.05)。结论MAGE-A1在RB组织中有一定量的表达且具有较强的特异性,提示有可能成为RB特异性免疫治疗的靶抗原。
Objective To investigate the expression of cancer-testis antigen(CTA) MAGE-A1 in tissues of retinoblastoma(RB) and the possibility of applying it as target antigen for specific immunotherapy for RB.Methods The expression of MAGE-A1 gene in tissues of RB,non-tumor retina and normal eye were detected with RT-PCR,and the expression of MAGE-A1 protein in tissues of RB and normal retina was analyzed by immunohistochemistry.Results The expressive rate of MAGE-A1 gene in RB tissues was 33.33%,but no MAGE-A1 gene expression was found in tissues of non-tumor retina and normal eyes,there was no significant correlation between the expression of MAGE-A1 gene and each clinical indicator(P〉0.05).The expressive rate of MAGE-A1 protein in RB tissues was 20%,but no MAGE-A1 protein expression was found in tissues of normal retina,the expression rate of MAGE-A1 protein was related with the age and children with or younger than 2 years old were more inclined to express(P〈0.05);There was high expression tendency in intraocular period(P〈0.01),and the expression of MAGE-A1 protein had no significant correlation with gender,tumor's differentiation and size(P〉0.05).Conclusions MAGE-A1 has a certain quantity expression in RB tissues with high specificity,which indicates that it can become the target antigen for specific immunotherapy for RB possibly.
