摘要
目的:建立并完善一种新的DNA(cDNA)检测方法,并探讨cDNA检测在鼻咽癌(nasopharyngealcarcinoma,NPC)患者诊断中的应用价值。方法:对196例经病理证实的NPC患者以微量基因组抽提试剂盒抽提血浆DNA,以荧光定量Realtime PCR测定含量,并以健康体检者125例作为对照组。结果:临床分期各组与治疗前相比(P均<0.05)差异有显著性;不论治疗前后,NPC患者各组与正常对照组相比,差异均有,(P均<0.05);cDNA含量与临床分期的关系:根据Ct值的逐渐减少,cDNA含量随着临床分期增加呈增高的趋势,但组间比较并无统计学意义(P>0.05)。结论:利用Realtime PCR法测定NPC患者cDNA,在NPC的诊断、疗效判断有着重要的临床应用价值。
Objective:To quantify the circulating DNA in plasma from patients of NPC and evaluate its potential clinical value. Methods : Plasma DNA was extracted by commercial "genomie DNA extraction kit" form 196 patients with NPC and 125 healthy individuals as control and detected by Realtime PCR method. Results:Significant difference (P 〈 0.05 ) in mean circulating plasma DNA levels between patients of NPC and healthy individuals, and it also has difference significant difference ( P 〈 0.05 ) between untreated patients and posttreatment in difference clinical stage with NPC. The level of circulating DNA was added gradually with the process of clinical stage. However, there was no significant different ( P 〉 0.05 ) between groups in difference clinical stage. Conclusions : Circulating plasma DNA detected by Realtime PCR has potential clinical value in diagnosis and therapeutic effect observing on patients of NPC.
