IVF受精失败卵子纺锤体、染色体的立体结构分析

摘要 目的:探索受精失败卵子纺锤体、染色体的立体结构变化及其相关因素。方法:选自本所受精卵子的材料分两组实验组:卵子来源于IVF取卵后24~40 h受精失败的MⅡ期卵子。对照组:为未授精卵子。两组卵子通过免疫荧光染片后,用Nikon激光共聚焦显微镜观察染色体、纺锤体的立体结构,根据年龄段、促排卵药的差别进行分组,用t和x^2检验进行统计学分析。结果:年龄小于30岁组的纺锤体、染色体正常率分别为:13.3%、13.3%,与对照组相比差异均无显著性,(P>0.05)。30~35岁组的纺锤体、染色体正常率分别为:16%,12%,纺锤体正常率与对照组差异相比,无显著性,(P>0.05),染色体正常率与对照组相比,差异有显著性,(P<0.05)。年龄大于35岁组的纺锤体、染色体正常率分别为:0%、0%,与对照组相比均差异有显著性,(P<0.05)。合计年龄小于35岁患者受精失败卵子的纺锤体、染色体正常率分别为:15%、12.5%,与对照组比,无显著性差异。果纳芬组的纺锤体及染色体的正常率均为12.5%,与对照相比,无显著性差异,(p>0.05)。丽申宝组纺锤体及染色体的正常率分别为11.1%,5.6%,纺锤体正常率与对照组相比,差异无显著性,(P>0.05),染色体正常率与对照组相比,差异有显著性,(P<0.05)。实验组的总的纺锤体和染色体正常率为12%、10%,与对照组相比均差异有显著性,(P<0.05)。结论:高龄妇女受精失败卵子的纺锤体和染色体的正常率下降;用丽申宝进行超促排卵的受精失败卵子染色体正常率下降;染色体非整体倍增加的一个相关因素,可能对纺锤体正常梭形结构的破坏。 Objective:To study the variation in the configurations of failured fertilization oocytes' spindle and chromosome and related factors. Methods: All of the failnred fertilization ooeytes were donated for this study from stimulated patients undergoing IVF treatment in our center. The oocytes which didn' t used to insemination were as the control group. After immunofluorescene staining, the study group oocytes and the control group oocytes were mounted on slides and observed under a Confocal Laser Scanning Microscope. The groups were divieded by different ages and stimulation medicine. We used t and χ^2 to analysis the data. Results : The patients age were 〈 30 years old, whose rates with normal spindle and chromosome configurations were 13% and 13%. There were no significant difference in the spindle and chromosome with normal configurations rates between the patients age 〈 30 years old group and control group. The rates of patients whose age were between 30 and 35 years old with normal spindle and chromosome configurations were 16% and 12%. There were no significant difference in the spindle with normal configurations rates between the patients age 30 - 35 years old group and control group. There were significant difference in the chromosome with normal configurations rates between the patients age 30 -35 years old group and control group. However, when the patients age were 〉 35 years old, the rate of the spindle and chromosome configurations were both zero. There were significant difference in the rates of spindle and chromosome with normal configurations between the patients age 〉 35 years old group and control group. The rates of spindle and chromosome with normal configurations of patients whose age were under 35 years old were 15% and 12.5% , respectively. There were no significant difference in the rates with normal spindle and chromosome configurations between the patients age under 35 years old group and control group. When the patients stimulated by the gonal-FSH, the rates of the spindle and chromosome configurations were both 12. 5% ,and there were no significant difference when eampared to the control group. When the patients stimulated by the liShenBao the rates of spindle and chromosome with normal configurations were 11.1% and 5.6%, and there were no significant difference in the spindle configurations rates between LiShenBao group and the control group. But there were significant difference in the rates of chromosome with normal configurations between LiShenBao group and the control group. The overall rates of the spindle and chromosome configurations were 12% and 10% in the study group. There were significant difference in the rates of the spindle and chromosome with normal configurations between the study group and the control group. Condutions:The rates of the spindle and chromosome with normal configurations were decreased in the elder women failured fertilization oocytes group. The rate of the chromosome with normal configurations was decreased in the the patients stimulated by the LiShenBao group. Our results indicated that the increasing aneuploidy rate should be correlated with the abnormal configurations of the spindle.
