摘要
目的:研究雷公藤红素对多发性骨髓瘤细胞系KMS12和KMS18的抑制作用及机理,以及对细胞周期的影响。方法:用CCK-8法研究抑制作用;凋亡检测采用V-Annexin-FITC和PI双染色法以及sub-S1法。流式细胞仪检测细胞周期。结果:雷公藤红素1μmol/L可抑制多发性骨髓瘤细胞系增殖,并诱导细胞凋亡。对细胞的S0期、S期、G2/M期均有影响。结论:雷公藤红素可引起多发性骨髓瘤细胞系凋亡并影响其细胞周期,具有潜在的临床应用价值。
Aim:To research the inhibition effect and the influence on cell cycle of the multiple myeloma cell lines,KMS12 and KMS18 by celastrol as well as the mechanism involved.Methods:The inhibition effect was measured with the CCK-8 kit,apoptosis was determined by staining with V-Annexin-FITCand PI labeling as well as the assay of sub-S1 phase.Cell cycle was determined by flow cytometer.Result :Cellular proliferation was inhibited by celastrol at low concentration undergoing apoptosis.Cell cycle was affected at S0 phase and S phase,as well as G2/M phase.Conclusion:Celastrol induced the apoptosis of MM cell lines in a relative low dose,thus has potential for clinic.
