雷公藤红素诱导多发性骨髓瘤细胞系凋亡及对其细胞周期的影响

Apoptosis induced by celastrol on multiple cell lines and the effect on cell cycle

目的:研究雷公藤红素对多发性骨髓瘤细胞系KMS12和KMS18的抑制作用及机理,以及对细胞周期的影响。方法:用CCK-8法研究抑制作用;凋亡检测采用V-Annexin-FITC和PI双染色法以及sub-S1法。流式细胞仪检测细胞周期。结果:雷公藤红素1μmol/L可抑制多发性骨髓瘤细胞系增殖,并诱导细胞凋亡。对细胞的S0期、S期、G2/M期均有影响。结论:雷公藤红素可引起多发性骨髓瘤细胞系凋亡并影响其细胞周期,具有潜在的临床应用价值。

Aim：To research the inhibition effect and the influence on cell cycle of the multiple myeloma cell lines,KMS12 and KMS18 by celastrol as well as the mechanism involved.Methods：The inhibition effect was measured with the CCK-8 kit,apoptosis was determined by staining with V-Annexin-FITCand PI labeling as well as the assay of sub-S1 phase.Cell cycle was determined by flow cytometer.Result ：Cellular proliferation was inhibited by celastrol at low concentration undergoing apoptosis.Cell cycle was affected at S0 phase and S phase,as well as G2/M phase.Conclusion：Celastrol induced the apoptosis of MM cell lines in a relative low dose,thus has potential for clinic.
