青藤碱影响HeLa细胞凋亡的COX-2信号通路的研究 被引量:3

The study of sinomenine on apoptosis in HeLa cell via COX-2 pathway

摘要 目的:研究青藤碱对宫颈癌HeLa细胞增殖的影响及其机制。方法:不同浓度青藤碱分别处理体外培养的人宫颈癌细胞系HeLa细胞,采用MTT法检测处理72h后HeLa细胞的增殖,流式细胞仪测定细胞凋亡。Westernblot检测COX-2和Bcl-2蛋白表达情况。结果:0.2、0.4mmol/L青藤碱处理HeLa细胞72h后,细胞增殖明显受到抑制;Westernblot分析表明,中高剂量组COX-2和Bcl-2蛋白表达水平下降;流式细胞仪细胞凋亡分析表明,各剂量组细胞凋亡率较对照组升高。结论:青藤碱在体外能抑制宫颈癌细胞生长,其机制可能与其抑制COX-2和Bcl-2蛋白表达,促进细胞凋亡相关。 Objective:To research sinomenine(SIN) intervention the proliferation of HeLa cell and induce apoptosis of tumour cell.Methods:HeLa cell were incubeded in vitro.This cell were divided into 3 experimental groups and 1 control group.The experimental group were given different dose SIN.The control group were given RPMI 1640.After incubated 72 hours,the proliferation of HeLa cell were detected by MTT and COX-2,Bcl-2 were investigated by Westernblot.To observe the general state of HeLa cell the Flow Cytometer were used to detect apoptosis of HeLa cell.Rseult:1.The proliferation of HeLa cell was inhibited after 72h with 0.2,0.4mmol/L SIN.2.The expression of COX-2 and Bcl-2 in middle and high dose experimental groups were decrease compare with control group(P0.05).3.The apoptosis rate were increased in experimental groups compare with control group(P0.05).Result:SIN can inhibit the proliferation of HeLa cell.The probably reason is SIN coult inhibit COX-2 and Bcl-2 expression.
