摘要
目的:探讨葛根素对糖尿病大鼠心肌的形态结构、功能及NF-κB表达的影响。方法:以45mg/kg剂量一次性尾静脉注射链脲佐菌素建立SD大鼠实验性糖尿病模型。应用葛根素(40mg/kg.d、80mg/kg.d)腹腔注射治疗8周后,测定各组大鼠血糖、血清LDH、CK含量及心肌组织SOD活性、MDA含量,检测心功能,应用光镜、电镜观察大鼠心肌形态结构,免疫组化方法观察心肌组织NF-κB表达情况。结果:与糖尿病模型组比较,葛根素治疗组大鼠血糖水平稍降,血清LDH、CK含量及心肌组织MDA含量显著降低,而高剂量组SOD活性明显增强,左心室功能、心肌形态结构尤其是超微结构明显改善,心肌组织NF-κB阳性表达明显减少。结论:葛根素对糖尿病大鼠心肌具有一定的保护作用,能明显改善心功能和心肌超微结构,可能与其减弱糖尿病大鼠心肌过氧化损伤及减少心肌组织NF-κB阳性表达有关。
Objective:To observe effects of puerarin on oxidative damage,ultrastructure and expression of NF-κB in cardiac tissue of diabetic rats.Methods:Diabetic rats were induced by a single intravenious injection of streptozocin(STZ).Animals were randomly divided into normal control group,diabetic model group,diabetic groups treated respectively with puerarin(40mg/kg·d and 80mg/kg·d)by peritoneal injection.8 weeks later,blood glucose,heart function,content of LDH and CK in serum,activity of SOD and content of MDA in myocardium were assayed.And the cardiac structure was observed with light microscope and transmission electron microscope.The expression of NF-κB in myocardial tissue was detected with immunohistochemistry method.Results:Compared with diabetic model group,in the rats treated with puerarin,blood glucose level,LDH and CK content in serum and content of MDA in cardiac tissue were decreased markedly,while activity of SOD in myocardium and heart function were enhanced.And the cardiac structure also was improved obviously,especially the ultrastructure.Expression of NF-κB in myocardial tissue was decreased significantly too.Conclusion:Puerarin could protect the myocardial tissue in rats against diabetes and improve heart function and myocardial ultrastructure,which may be associated with lessening oxidative damage and expresson of NF-κB in myocardial tissue.
