摘要
目的:探讨丹参多酚酸盐治疗动脉粥样硬化的可能分子靶标。方法:将单核巨噬细胞U937设为对照组、模型组、药物处理组。其中,模型组、药物处理组以ox-LDL刺激1h,再于药物处理组中加入丹参多酚酸盐。待药物作用细胞3h后,RT-PCR法检测巨噬细胞Resistin、SRA、CD36mRNA表达水平。结果:与对照组相比,模型组Resistin、SRA mRNA表达水平均增高(P<0.01),药物处理组Resistin、SRA mRNA表达水平无差异(P>0.05);与模型组相比,药物处理组Resistin、SRA mRNA表达水平均降低(P<0.05)。各组细胞CD36 mRNA表达未见显著变化。且丹参多酚酸盐对抵抗素表达的影响作用与对清道夫受体A表达的影响作用呈正相关。结论:丹参多酚酸盐可能以抵抗素作为其药理作用的分子靶标之一,且对抵抗素表达的影响可能与清道夫受体A的表达受抑制有关。
Objective:To investigate the maybe molecule target which salvianolate is how to treat the artherosclerosis(AS).Methods:The U937 monocyte-derived macrophages were separated to three groups:control group,model group and treated group.The model group and treated group were stimulated by ox-LDL.After 1h we added the salvianolate to treated group.Three hours later,we detected the mRNA expression of resistin,SRA and CD36 in macrophages by RT-PCR.Results:Compared with control group,model group significantly increased the mRNA expression of resistin and SRA(P0.01),but treated group didn't change the mRNA expression of the two(P0.05);Compared with model group,treated group significantly degraded the mRNA expression of resistin and SRA(P0.05).There was no change in mRNA expression of CD36.And there was a positive correlation that the effects of salvianolate on expression of resistin and SRA.Conclusion:Resistin maybe one of the molecule targets in pharmacological action of salvianolate.And the effects of salvianolate on expression of Resistin is associated with the inhibited expression of SRA.
