摘要 目的:研究胃癌组织中雷帕霉素靶蛋白(mTOR)、缺氧诱导因子1α(HIF-1α)的表达及微血管密度变化,探讨mTOR、HIF-1α在胃癌发生发展中的作用及与血管生成的关系。方法:免疫组织化学法检测70例胃癌组织及45例癌旁正常组织中mTOR和HIF-1α蛋白的表达,以CD34标记血管内皮计数微血管密度(MVD)。结果:70例胃癌组织中mTOR呈阳性表达36例,占51.4%,HIF-1α阳性表达47例,占67.1%,均显著高于癌旁组织中的10.7%和17.8%(χ2分别为19.408和26.751,P均<0.01);mTOR与HIF-1α的表达呈正相关(r=0.157,P<0.05);mTOR、HIF-1α表达与肿瘤的TNM分期、浸润深度、淋巴结转移、远处转移显著相关(P<0.05)。胃癌组织的MVD较癌旁正常组织显著增高(t=4.358,P<0.05)。mTOR、HIF-1α均阳性者MVD显著高于mTOR、HIF-1α均阴性者(F=23.090,P<0.01)。结论:mTOR及HIF-1α在胃癌组织中呈阳性表达,胃癌组织的微血管密度显著高于癌旁组织。 Objectives: To detect the expressions of mTOR and HIF-1αand their relationship with angiogenesis in gastric carcinomas. Methods: The expressions of mTOR and HIF-1αin gastric carcinomas tissues(n=70)and normal tissues(n=45)were measured by immunohistochemistry staining.CD34 immunostaining was used to measure the microvascular density(MVD).Results: The rates of the mTOR and HIF-1α positive expression were 51.4% and 67.1% respectively. MVD value of gastric cancer tissues was much higher than that of normal tissues(t=4.358, P〈0.05). MVD values were much higher in tumor tissues of mTOR (+)/HIF-1α (+) than those of mTOR(-) /HIF-1α(-)(F=23.09, P〈0.05). Overexpressions of mTOR and HIF-1α were significantly associated with TNM stage, invasion depth, lymphnode metastasis or distant metastases(P〈0.05).Significant correlation was seen between mTOR and HIF-1α positive expressions (r=0.157, P〈0.05). Conclusion: mTOR and HIF-1αare indistinctively overexpressed in gastric carcinoma, and may play an important role in the angiogenesis and carcinogenesis.
