摘要
目的建立黄芪及相关药材的高效液相-蒸发光散射检测器(HPLC-ELSD)色谱指纹图谱的评价方法。方法确定实验条件并进行方法学考察,色谱条件:依利特HypersilODS2色谱柱(4.6mm×250mm,5μm);流动相:A相为乙腈,B相为水,梯度洗脱;流速1.0ml/min;柱温为室温;进样量20μl。ELSD检测器:漂移管温度40℃,载气(空气)压力3.5Bar,增益值:7。采用国家药典委员会颁发的《中药色谱指纹图谱相似度评价系统A版》软件进行指纹图谱分析。结果黄芪药材HPLC-ELSD色谱指纹图谱具有较好的稳定性、精密度和重复性,相对保留时间和相对峰面积的RSD均小于5.0%,标定8个共有峰,相似度计算结果均大于0.85,可以同多序岩黄芪和梭果黄芪进行有效区分。结论本研究将有助于加强黄芪药材的质量控制,能够有效的区分黄芪药材同其他相关样品。
Objective To establish evaluation methods of Radix astragali and the relative herbs with HPLC-ELSD chromatographic fingerprints. Methods The conditions of experiment were confirmed and the validation of the method was also tested. The chromatography conditions were defined as Hypersil ODS2 column (4.6 mm ×250 mm, 5 μm). mobile phase was A :ACN, B :H2O, gradient elution, flow speed was 1.0 ml/min, temperature of column was room temperature, injection volume: 20 μl. The ELSD conditions were as follows: the temperature of drift tube was 40℃ , the gas pressure was 3.5 Bar, the value of gain was 7. The chromatographic fingerprints were analyzed with similarity evaluation system for chromatographic fingerprint of TCM (A Edition ) issued by the National Pharmacopoeia Committee. Results HPLC-ELSD chromatographic fingerprints of Radix astragali showed better stability, precision and reproducibility, RSD of relative retention of times and peak areas were less than 5.0% , 8 common peaks were demarcated and the results of similarity calculation were more than 0.85. Furthermore, the results of similarity calculation can distinguish Hedysarum polybotrys Hand. -Mazz. and Astragalus ernestii Comb effectively. Conclusion The study may contribute to in controlling the quality of Radix astragall and also can effectively distinguish Radix astragali from other samples.
