葡萄品种资源的SSR鉴定及遗传多样性分析 被引量:20

SSR identification and analysis of genetic diversity of grape cultivars

摘要 用筛选出的12对SSR引物分析了39个葡萄品种的遗传多样性。结果表明:12对引物在上述品种中共扩增出155条谱带,其中多态性带149条,占扩增总带数的96.13%,特异性条带8条,占5%,所扩增的片段大小在50-1 000 bp之间,不同引物扩增的带数在5-24之间,平均12.9条。扩增条带最多的是引物VrZAG67,扩增出24条带 最少的是VrZAG47,仅扩增了5条带。品种间遗传相似系数变异在0.374-0.974之间,在相似系数为0.660的阈值下,可将39份材料分为3大类群。第1类包括“格拉卡”和“美人指”2个品种;第2类包括“玫瑰香”、“红旗特早”、“达米娜”、“红玫瑰”等30个品种;第3类包括“京云”、“奥古斯特”、“无核早红”、“巨峰”等7个品种。 Twelve pairs of SSR primers were used to study genetic diversity among 39 grape varieties. The result showed that 155 bands were amplified and the polymorphic percentage was 96.13%. There are 8 special bands with the percentage of 5%. The number of amplified bands per primer pair ranges from 5 to 24, with an average of 12.9. With the largest number, primer VrZAG67 amplified 24 bands, and the least was 5 by VrZAG47. The genetic similarity coefficient among all grape varieties ranges from 0. 374 to 0. 974. Cluster analysis was made with unweighted pair group method for arithmetic averages by using genetic similarity coefficient. At the genetic similarity of 0. 660, 39 materials are clustered into 3 groups. The first group consists of "Greaca" and "Manicure Finger". The second group consists of 30 varieties, including "Muscat Hamburg", "Hongqitezao", "Tamina", and "Hongmeigui". The third one is composed of 7 varieties, including "Jingyun", "Augusta", "Wuhezaohong", and "Jufeng".
