摘要
为了探讨中国猪群TTV的感染现状以及是否存在新的基因亚型,基于TTV1和TTV2相对保守的5’非翻译区(5’UTR)设计引物,采用PCR法对2008~2009年采集于中国江苏省的138份猪血清进行了检测。结果表明,中国猪群,TTV1的感染率为15.9%,TTV2的为34.8%,共感染率为5.8%。所扩增片断的核苷酸序列分析结果表明,中国TTV1和TTV2流行毒株之间的核苷酸同源性分别为87.3%~93.8%和74.8%~99.1%,与GenBank其他TTV1和TTV2毒株的同源性分别为69.6%~100.0%和74.8%~99.1%。系统进化分析结果表明,中国TTV1和TTV2流行毒株存在新的基因亚型。
To investigate the genomic structural characteristics of swine Torque teno viruses from China and their prevalence,two pairs of primers were designed based on conserved nucleotide of 5' untranslated regions of swine Torque teno virus genogroups 1 and 2. Of 138 swine serum samples from different Jiangsu herds in 2008 and 2009 ,TTV1 DNA was detected in 22 samples( 15.9% ),TTV2 in 48 samples (34. 8% ) and both in 8 samples(5.8% ). Sequence analysis revealed that the Chinese TTV1 and TTV2 isolates shared 87. 3%-93. 8% and 74. 8%-99. 1% nucleotide sequence identity,respectively,and 69. 6%-100. 0% and 74. 8% -99. 1% with other TTV1 and TTV2 isolates submitted to GenBank, respectively. Phylogenetic analysis demonstrated that there existed the novel genetic subtypes among Chinese TTV1 and TTV2 groups.
