摘要
人的SLP-2基因是一个新的肿瘤相关基因,它在多个癌组织里高表达,如食管鳞状细胞癌组织和肺癌组织。其中在乳腺癌组织的高表达和病人的存活率负相关,这说明该基因很可能在乳腺癌的发生中发挥重要作用。为了研究SLP-2基因在肿瘤里的功能,将人类SLP-2基因全长编码区定向连入pEGFP-C3质粒,使SLP-2蛋白可以与绿色荧光蛋白在乳腺癌细胞MCF-7内融合表达。而且细胞荧光实验发现SLP-2蛋白主要定位在MCF-7细胞的胞质内,这为进一步研究SLP-2基因在乳腺癌中的功能奠定了实验基础。
Human stomatin like protein 2 ( SLP-2 ) is a novel cancer-related gene. Its protein is overexpressed in many human cancers, such as esophageal squamous cell carcinoma and lung cancer. In Breast cancer, high-level SLP-2 expression were associated with significantly decreased survival. To study the function of SLP-2 in these cancers, the coding region of human SLP-2 gene was successfully subcloned into the eukaryotic green fluorescent protein(GFP) expressing recombinant plasmids, pEGFP-C3. The vector was then transfected into MCF-7 cells for efficient expression. Moreover, results from fluorescent observation shows that GFP-SLP-2 mainly localized in cytoplasma, thus providing a useful clue for further research on the function of SLP-2 in breast cancer.
