脑康Ⅱ号改善糖尿病大鼠认知障碍的研究 被引量:2

Research of Naokang Ⅱ in improving diabetic rat's cognitive impairment

摘要 目的:研究脑康Ⅱ号对糖尿病大鼠学习记忆功能的干预情况。方法:使用脑康Ⅱ号对糖尿病大鼠进行干预,采用水迷宫实验和流式细胞技术,分别观察糖尿病大鼠的行为学改变和海马神经细胞凋亡发生率的变化,并用免疫组化法检测海马CA1区凋亡相关蛋白半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)、B细胞淋巴瘤/白血病-2基因(Bcl-2)的表达水平。结果:脑康Ⅱ号能够显著缩短大鼠Morris水迷宫到达站台时间,并减少大鼠海马神经细胞凋亡率,上调海马CA1区Bcl-2表达,减少Caspase-3表达。结论:脑康Ⅱ号能够通过上调Bcl-2蛋白、下调Caspase-3蛋白的表达,减少海马神经细胞凋亡,改善糖尿病大鼠认知障碍。 Objective: To research the improvement of Naokang Ⅱ in treating diabetic rat's cognitive impairment. Methods: Naokang Ⅱ was used to treat diabetes. The change of rat's ability of learning and memory function was tested by water maze. The percentage of neuron apoptosis was detected through flow cytometry. The expression of Caspase-3 and Bcl-2 proteins in hippocampal CA1 region was measured with immunohistochemistry. Results: Naokang Ⅱ could significantly reduce the swimming time to platform and the percentage of neuron apoptosis, and increase the expression of Bcl-2 and decrease the expression of Caspase-3 in hippocampal CA1 region of model rats. Conclusion: Naokang Ⅱ could improve diabetic rat's cognitive impairment by reducing hippocampus neuron apoptosis and increasing the expression of Bcl-2 and reducing the expression of Caspase-3 in the hippocampus.
