消敏颗粒抗Ⅰ型超敏反应作用机制的实验研究 被引量:3

摘要 目的通过观察消敏颗粒对小鼠耳异种被动皮肤超敏反应及对大鼠肥大细胞释放组胺、5-羟色胺的影响,进一步探讨消敏颗粒抗Ⅰ型超敏反应作用机制。方法检测耳异种被动皮肤超敏反应(PCA)小鼠耳A值;并对Ⅰ型超敏反应致敏动物模型大鼠腹腔肥大细胞运用荧光测定改良法测定组胺释放率;免疫组化染色SABC法检测5-羟色胺的积分。结果消敏颗粒各组PCA均明显减轻,与实验对照组相比差异有统计学意义(P<0.01);消敏颗粒各组大鼠腹腔肥大细胞组胺的释放量均明显降低,与实验对照组相比差异有统计学意义(P<0.01)。消敏颗粒各组部分腹腔肥大细胞颗粒脱失,5-羟色胺表达减弱。对细胞进行综合积分可见,消敏颗粒各组均明显高于实验对照组,差异有统计学意义(P<0.01)。结论消敏颗粒能显著抑制小鼠异种皮肤被动过敏反应;消敏颗粒可通过减少腹腔肥大细胞脱颗粒,抑制其释放组胺及5-羟色胺,发挥抗Ⅰ型超敏反应作用。 Objective To observe the effect of Xiaomin granules on passive cutaneous anaphylaxis(PCA) mice ears,as well as the infection on histamine and 5-HT released from mast cells of the rats,and to investigate the effect and mechanism of Xiaomin granules on typeⅠhypersensitivity.Methods The A of PCA of mice ears had been tested.Fluorometric assay was used to measure the histamine rate in the mast cells in abdominal cavity of the rats model caused by typeⅠhypersensitivity.The method immunohistochemical SABC was used to investigate the integral of 5-HT.Results The result indicated that PCA of all Xiaomin granules groups was obviously reduced compared with experiment contradistinguish group. There is statistical significance(P0.01).The releasing histamine of mast cells in abdominal cavity of the rats in Xiaomin granules groups was obviously reduced compared with experiment contradistinguish group.There was statistical significance (P0.01).The peritoneal mastocyte of all Xiaomin granules groups had escaped pellets and the expression of 5-HT was weaken.All groups of Xiaomin granules had statistical significance compared with experiment contradistinguish group(P0.01).Conclusion Xiaomin granules can inhibit passive cutaneous anaphylaxis ears of the mice obviously.Xiaomin Granules can reduce mast cells degranulation,inhibiting the release of histamine and 5-hydroxy tryptamine,then takes effect on the mechanism of Xiaomin granules on typeⅠhypersensitivity.
